摘要:

Endogenous metabolism and environmental exposure can both give rise to DNA damage.  Organisms are evolved with various DNA repair pathways to remove damaged nucleosides from their genomic DNA, and unrepaired DNA lesions may result in genomic instability by inhibiting DNA replication and transcription, and inducing mutations in these processes.  In this presentation, I will discuss our recent development and applications of integrated chemistry and biology approaches, which include mass spectrometry-based bioanalytical chemistry, synthetic organic chemistry, molecular biology and genetics, to assess the occurrence and repair of DNA lesions.  In addition, we will show that this interdisciplinary approach can provide a molecular level understanding about the degrees to which these DNA lesions perturb the transmission of genetic information by diminishing the efficiencies and fidelities of DNA replication and transcription.  Particular emphasis will be placed on DNA damage products arising from endogenous oxidative stress and from exposure to alkylating agents.
