摘要
O-GlcNAc transferase (OGT) is an essential human enzyme that catalyzes the addition of a single N-acetylglucosamine to serine or threonine residues of intracellular proteins. This unique post-translational modification, named O-GlcNAcylation, modulates a wide range of signaling pathways linked to glucose metabolism and its abnormality has been linked to many human diseases. In this talk, I will discuss recent results on the molecular mechanism of O-GlcNAcylation, a new class of OGT inhibitors with an unprecedented mechanism of inhibition, and the novel proteolytic activity of OGT in the maturation process of Host Cell Factor 1 (HCF-1) an important regulator of the cell cycle. These studies facilitate rational design of chemical tools to dissect the biological functions of OGT and O-GlcNAcylation.
